I want to Improve Separation Resolution

It's difficult to achieve high separation resolution from slab gels. Though high resolution gels exist, the human factor often leads to inconsistencies, because one technician might use slightly different gel preparation conditions compared to another technician. These differences contribute to unwanted run- to-run variability. So, labs are motivated by two factors in switching from manual gels to an analytical instrument:

  • they want more consistent results, and
  • they want better separation resolution of PCR amplicons generated from microsatellites.

The AdvanCE™ FS Nucleic Acids Analyzer is far superior to traditional slab gels in both regards. Resolution levels as low as 2bp are achieved on the AdvanCE™ FS Nucleic Acids Analyzer. And four gel formulas allow labs with the AdvanCE™ FS Nucleic Acids Analyzer to look at a variety of fragment sizes – small, medium, large and genomic. The gels are:

Gel Type Capabilities
DNF-900 Best for small DNA fragment analysis, 35 - 500 bp.
Gel resolution of 2-5 bp
DNF-910 Broad range DNA fragment gel; analyze fragments 35 – 1,500 bp
Gel resolution varies from 5bp <500bp, 5-10bp >500bp, 20bp >1000 and 50bp <2000
DNF-920 Medium and large DNA fragment analysis, 75 - 15,000 bp.
Gel is also capable of separating major plasmid DNA species, supercoiled, relaxed and linear species.
DNF-930 Large DNA fragments, 75 – 20,000

Many factors can impact the consistency of resolution; the following factors have been shown to improve separation resolution on the AdvanCE™ FS Nucleic Acids Analyzer:

  1. QC performance tested reagents
  2. Fresh gel replacement in every capillary prior to each run
  3. Automated injection of samples under precisely controlled injection conditions
